Studies on the biochemistry of streptomyces. III. Glucose catabolism in Streptomyces griseus.

The operation of the tricarboxylic acid cycle and fixation of CO2 in Streptomyces griseus has recently been demonstrated in a series of tracer studies, Gilmour et al. (1955); Butterworth et al. (1955). As to the nature of the pathways of glucose catabolism in this organism, the operation of the Embden-Meyerhof glycolytic scheme and the direct oxidative pathway were indicated by inhibition studies carried out by Hockenhull et al. (1954). Cochrane et al. (1953) reported preferential decarboxylation of the C-1 of glucose by Streptomyces coelicolor and Streptomyces reticuli, and concluded that a direct oxidative pathway is operative in these two organisms. In the present work, time course experiments on the utilization of C14 specifically labeled glucose and acetate by S. griseus have been carried out. The results point to the operation of glycolysis in conjunction with the tricarboxylic acid cycle as the major catabolic pathways of glucose catabolism in this organism. A minor portion of glucose was metabolized by way of phosphogluconate decarboxylation. The change in catabolic pattern of glucose among cultures of different ages is discussed.
